NNC-711 is a novel research chemical anticonvulsant and GABA reuptake inhibitor that produces net effect of GABA in a particular area under different physiologic conditions. It was studied the effect of NNC-711 on spontaneous synaptic currents in cultured hippocampal neurons to investigate its physiologic mechanism of action. NNC-711 have potent anticonvulsant activity against pentylenetetrazol- and audiogenically induces seizures in rodents. A GABA uptake inhibitor was expected to increase either IPSC amplitude or duration as its mechanism of action if either of these properties were affected by clearance of GABA from the synaptic cleft. On the contrary, NNC-711 decreased the amplitude of IPSC dose dependently. Exogenously applied GABA-induced current was not changed by NNC-711, indicating that NNC-711 did not directly block postsynaptic GABAA receptors. NNC-711 also inhibites EPSC amplitude. Abolished the effects of NNC-711 on both IPSCs and EPSCs.
